Topical Fat ?Reduction Effects of Mesotherapy: Randomized Controlled Trial

Abstract
Background: Localized deposits of excess adipose tissue may not be medically dangerous, but can be psychologically distressing to women. Mesotherapy has been widely used for cosmetic purposes. Few studies have been published in the English?language literature on the safety, efficacy, and action mechanism of mesotherapy when performed for body contouring. This study investigated the effects of mesotherapy on topical fat?reduction in the abdominal region.

Methods: A single-center, double-blind, randomized placebo-controlled trial was performed. Fifty-six pre-menopausal healthy female volunteers fulfilled the requirements for the study. The formulations for the intervention group were composed of procaine, caffeine, pentoxifylline, vitamin C, and normal saline. The formulation for the placebo group was 10 ml of normal saline. The treatments consisted of 8 sessions during 8 consecutive weeks. The treatment protocols were multiple superficial injections using ¡¯point-by-point¡¯ injection techniques with 30 gauge x 13mm needles around the abdomen. Fat mass reductions were evaluated according to changes of body weight, % body fat, waist circumference, sagittal anterior diameter(SAD), abdominal subcutaneous fat area, and anterior subcutaneous fat thickness by single slice fat CT scan.

Results: The intervention group did not show any significant changes in all of the body measurements compared to the placebo group. There were no serious complications found during the study. The most frequent adverse effect was bruising around the injection site, which was found in 53.6% of the intervention group and in 10.7% of the placebo
group(P=0.001). Other adverse effects were mild and not significantly different.

Conclusions: Mesotherapy using medications did not significantly reduce topical abdominal fat deposits compared with mesotherapy without medications. A careful consideration is needed till the topical fat reduction effect of mesotherapy is confirmed.
